Abstract


Aeschynanthus radicans var. ‘Monalisa’ dikenal dengan nama bunga lipstik, yang merupakan marga epifit dari suku Gesneriaceae. Tanaman ini mengalami dikogami jenis protandri. Pengetahuan mengenai viabilitas serbuk sari dari tanaman dikogami sangat diperlukan untuk menunjang keberhasilan persilangan. Penelitian ini bertujuan untuk menentukan tahap-tahap perkembangan morfologi bunga A. radicans var. ‘Monalisa’, viabilitas serbuk sari dengan uji pengecambahan in vitro dan uji pewarnaan, serta mempelajari korelasi viabilitas serbuk sari antara uji pengecambahan in vitro dengan uji pewarnaan. Tahapan dari inisiasi tunas bungahingga mencapai antesisadalah 34-35 hari dan rata-rata masa gugur pada stadia H+12 dan H+13. Hasil penelitian pendahuluan pada stadia bunga H0 (antesis) memperoleh waktu optimum pengecambahan serbuk sari yaitu 8 jam untuk serbuk sari dari tangkai sari panjang dan 9 jam untuk serbuk sari dari tangkai sari pendek. Viabilitas serbuk sari tertinggi dalam media BK dijumpai pada stadia H+2 dari tangkai sari panjang dan tangkai sari pendek masing-masing yaitu 55.7% dan 56.7%. Uji pewarnaan dengan viabilitas serbuk sari tertinggi dijumpai pada stadia H+1 dari tangkai benang sari panjang dan tangkai benang sari pendek masing-masing 41.8% dan 48.0% dengan anilin blue 1%; serta 29.4% dan 27.2% dengan I2KI 1%. Viabilitas serbuk sari pada media BK dengan pewarna anilin blue 1% dan I2KI 1% berkorelasi positif.


Aeschynanthus radicans var. ‘Monalisa’ also known as lipstick flowers, are epiphytic genus of the Gesneriaceae family. These plants experienced the protandri types of dichogamy. Knowledge of pollen viability from dichogamy plants is needed to increase  the success rate of a crossover. This research aimed to determine the developmental stage of flower morphology A. radicans var. ‘Monalisa’, pollen viability with in vitro germination test and staining test, and the correlation of pollen viability between in vitro germination test and staining test. Length of the process from flower bud initiation until the formation of anthesis was 34-35 days and the average abscission period took place at stadia H+12 and H+13. The result of the preliminary study at the stadia H0 (anthesis) showed that pollen germination obtained its optimum time within 8 hours for the long filaments pollen and took 9 hours for the short filaments pollen. The highest pollen viability in BK media found at stadia H+2, which had the value for long filaments and short filamens 55.7% and 56.7% respectively. Staining test with the highest pollen viability found at the stadia H+1, with the value of long filaments and short filaments 41.8% and 48.0% respectively with 1% aniline blue; and 29.4% and 27.2% respectively with 1% I2KI. The pollen viability in BK media with 1% aniline blue and 1% I2KI were positively correlated.
