ORGANOGENESIS TUNAS SECARA LANGSUNG PADA PAMELO (Citrus maxima (Burm.) Merr.)

Abstract


Konservasi in vitro pamelo (Citrus maxima (Burm.) Merr.) memerlukan tunas in vitro sebagai eksplan. Tunas in vitro diperlukan dalam konservasi untuk mempermudah pemulihan dan penggunaannya setelah konservasi. Dua percobaan dilakukan untuk mendapatkan eksplan dan media yang efektif untuk memperoleh tunas in vitro pamelo secara langsung. Eksplan daun, akar dan epikotil diperoleh dari kecambah in vitro pamelo ‘Adas Duku’. Percobaan pertama dilakukan untuk mendapatkan media mengandung sitokinin dan auksin yang efektif dalam menginduksi tunas adventif. Percobaan disusun dengan rancangan acak lengkap. Media MS mengandung kombinasi BAP (0; 1; 2 ppm) dan NAA (0; 0,5; 1 ppm) digunakan sebagai perlakuan. Percobaan kedua dilakukan pada eksplan epikotil pamelo untuk mengetahui pengaruh posisi kultur pada pembentukan tunas. Hasil pengamatan menunjukkan bahwa tunas dapat terbentuk secara langsung pada eksplan daun, akar dan epikotil. Eksplan daun hanya berespon di ruang gelap, yaitu sebanyak 5,55% dapat membentuk 1 tunas/eksplan di media MS + BAP 1 ppm. Eksplan akar hanya berespon di ruang terang, yaitu sebanyak 60% dapat membentuk 1 tunas/eksplan di media MS0. Eksplan epikotil yang dikultur horisontal dapat membentuk tunas secara langsung di ruang terang pada media MS0, yaitu sebanyak 30% dapat membentuk 1–2 tunas/eksplan, sedangkan di ruang gelap tunas terbentuk secara langsung di media MS0 dan MS + BAP 1 ppm, namun dengan penampilan tunas yang lemah karena etiolasi. Epikotil yang dikultur secara vertikal di ruang terang sebanyak 100% dapat menghasilkan 1–3 tunas/eksplan.
